I have access to both .com and .co.uk domains, and I am thinking of trying the Amazon affiliate route. Should I focus on UK sales on a UK site, or is Amazon.com t
A .co.uk domain is more likely to rank well on http://www.google.co.uk/ncr so you should primarily be targeting a UK audience with that, whilst a .com will rank better on http://www.google.com/ncr and may rank less well on http://www.google.co.uk/ncr as well.
However, you'll need to sign up to both Amazon co.uk and/or .com to earn from the relative countries.
