You can use the 'title="meaning or description"' parameter in the <span> tag,
Example: using the HTML code to have a popup meaning for the words 'Heaps Good!' would be written like this:

<p>
This is <span style="color:#66c;" title="Australian term meaning extremely wonderful, or exciting">heaps good!</span> check it out!
</p>

You can copy and paste this code into your page to test it out and see what it looks like. Basiclly it looks like this:
This is heaps good! check it out
the words 'heaps good!' are in blue text, and when you mouse over it, a box pops up with "Australian term meaning extremely wonderful, or exciting"
